Comparison of yaxin recombinant aprotinin (RTI16) and sigma extract aprotinin (A1153)
OBJECTIVE: To compare the similarities and differences of specific activity, absorbance, acidity and purity between Yaxin Recombinant Aprotinin (Cat. No. RTI16, Lot No.: RTI161101) and sigma Aprotinin (Cat. No. A1153, Lot No.: SLBP2656V).
experimental method:
1. Determination of protein content, specific activity, absorbance, acidity test. Weigh a certain amount of powder of two enzymes, dissolve them in pure water, prepare 2.5g/ml enzyme solution, and determine the protein content according to the absorption coefficient method. 280 nm , 1 cm (1 mg/ml) = 0.84, activity assay was performed according to SOP and specific activity was calculated. The enzyme solution was diluted to 3EPU/ml, and the absorbance was measured at 277 nm, and the acidity of the enzyme solution was determined to be not more than 0.8.
2. Determination of HPLC purity 1 The molecular sieve analysis column was selected as TSK-GEL G2000SWXL.

Experimental results:
The protein content, specific activity, absorbance and acidity of the two proteins were basically the same. The specific activity of the recombinant aprotinin (RTI161101) was slightly higher than that of sigma aprotinin (A1153).
